What is a nasopharyngeal swab?

A nasopharyngeal swab test is different from a nasal swab test. Nasopharyngeal swabs are used to collect a sample from the nasopharynx, the upper part of the throat behind the nose. Because of this sample collection location, patient's may experience discomfort during the swab test. Nasopharyngeal testing should only be done by nurses or other healthcare professionals. At home nasopharyngeal testing is not recommended, although nasopharyngeal testing is not considered painful and patients should only experience temporary discomfort.

How is a nasopharyngeal swab test different from a nasal swab test?

A nasal swab test collects a sample from the nasal membrane, about an inch in the nose. A healthcare worker will use a medium-tip swab that can be made from flocked fibers, polyester, or medical grade foam to swab the inside of the patient's nose in a circular motion for 10-15 seconds, ensuring the swab tip makes contact with the nasal wall. Once one nostril is swabbed, then the health care professional will do the same procedure on the other nostril using the same swab. Nasal swab tests are easy to administer for both patient's and nurses. Nasal swab tests can be performed by individuals at home.

A nasopharyngeal swab test collects a sample from the upper part of the throat in the nasopharynx region. Because of the invasive nature of this test and the risk of patient discomfort, a mini-tip swab should be used. The mini-tip swab is generally a type of flocked material with a thin polystyrene shaft that offers a little flexibility. Once the swab makes contact with a nasopharynx region, the healthcare worker should rotate the swab a few times and then gently remove the swab.¹ If the nurse or doctor determines that enough sample has been collected, there is no reason to perform the same procedure on the other nostril. Nasopharyngeal testing should only be performed by health care professionals with proper training. Nasopharyngeal testing should not be performed at home due to risk of pain, discomfort, and improper collecting methods leading to improper test results.
